After the error occurred, I did make sure that the login that does the backup had read/write permissions on everything, but that fix only lasted for one backup.

As far as I know, I've changed nothing.

That particular computer does only one thing which is to run a scheduled backup once a day. It is a computer that is located in another town & is my offsite backup ...
