Excluding "C:\Users\...\AppData\Local\Microsoft\..." does not work. How do I get this to work or how do I get the error message eliminated?

As clearly stated in the FFS Manual page on File Exclusion, first bullet, Include and Exclude item entries must be relative to the left and right base directories.
You are using an absolute location.

Best way for filter setup is to run a compare, find what you want to exclude in the results, and then right-click it to filter for your needs. Syntax is guaranteed to work

OK.... But a logical and rational methodology would be far more useful and usable. EXCLUDE and INCLUDE should be in reference to the source folder/directory. However, I will try the right-click method and hope it works as described. Thanks.

That may make sense for a Mirror or Update sync where there is a clear source folder, but obviously very much not so for Two-way or Custom syncs. And having two different ways of specifying In- and Excludes, depending on the sync variant being run, makes even less sense.
So, it is more logical and rational, and at least as useful and useable, to use the method used presently in FFS.
